- Description: With global competition becoming a key concept in the majority of companies worldwide, it is necessary for organisations to develop operations strategies that enable them to meet their customers‘ needs. Organisations will never be capable of reaching future goals without their customers. For this reason it is imperative for organisations to identify what competitive factors are required by the market, and which performance objectives are needed in order to increase the competitiveness of the organisation. In this research, Chapters 1 and 2 were used to identify the main and subproblems of the research, to delimit the research and to provide literature background to operations strategies and related operations topics. The empirical study in Chapter 3 was used to gather information pertaining to the specific requirements of ESA’s customers, and the operations performanceof ESA. Chapter 4 was used to analyse the data gathered from the empirical study. From this analysis it was possible for the researcher to identify the specific requirements of ESA’s customers and the operations performance of ESA. Chapter 5 was used to provide recommendations to bridge the performance gap between the customer requirements and the operations performance of ESA. By effectively implementing all of the recommendations suggested in this research, ESA will be able to improve its operation performance and meet the specific requirements of its market and its customers.

Because of high competition in this industry, it is of vital importance not to be out-marketed by competitors by keeping abreast of new trends in promotional strategies. In order to obtain more relevant information about promotional strategies a literature study was conducted to develop a questionnaire to survey the importance of promotional strategies and determine which promotional strategies are being used by the FET Colleges. The East-London, Queenstown and Ezibeleni Public FET Colleges form the respondents. From each college five academic staff, two management members and five students formed the respondents. Community members, business community members, the Department of Education and the Department of Labour representatives were also included in the sample from each of the selected FET Colleges. The findings of the survey revealed the high level of agreement in terms of the importance of the promotional strategies identified in the literature study with the exception of some promotional strategies, which were identified as not compatible with the FET target market. The study recommended that further research be done on a larger sample size focusing on particular stakeholders eg. students of the FET Colleges in a specific area.

- Description: Red-billed Quelea Quelea quelea have expanded their range in the Eastern Cape and now occur throughout the year in new areas. Recent changes in agricultural practice have probably been a contributing factor as flocks are larger than previously recorded and were more often associated with artificial food sources than natural grasses. Ringing and census data indicate that quelea have reduced movements during the non-breeding season and may display strong fidelity (over successive years) to dry season quarters with reliable food supplies. Although the recapture/recovery rate for quelea in the Eastern Cape (1.0-2.5%) was higher than the national average, it was still lower than expected and there may be considerable movement between nearby feeding sites. Some quelea breed locally, but most disperse during summer and their numbers at the feedlots were generally highest in winter and spring. The breeding season of quelea is later than other ploceids in the region and post-nuptial moult overlaps with winter. Replacement of primary wing feathers is relatively slow (124 days), and this is considered an adaptation to minimise disruption of flight capabilities and insulation. Significantly more quelea in the Eastern Cape have breeding plumage suffused with pink than in other southern African populations. However, during the non-breeding season there is apparently considerable intermixing between local populations and those from further north and the existence of a local sub-species is not supported. Seeds of two grass species, Echinochloa sp. and Urochloa panicoides, and two weed species, Amaranthus sp. and Chenopodium sp., were important in the diet of both quelea and Laughing Doves Streptopelia senegalensis. Maize comprised a large proportion of the diet of these species and losses at one ostrich feedlot were estimated at over R 17 000 in two years. Dependence on artificial food sources was generally greatest in winter and spring, but economically significant damage was not confined to this period. Alpha-chloralose showed good potential for reducing numbers of problem birds at livestock feedlots. However, the dynamic nature of problem bird populations favours a non-lethal management approach. Reduction of feed loss through manipulation of the ostrich ration could provide a relatively cheap and effective alternative to lethal control if applied appropriately.

The ecophysiology of selected coastal dune pioneer plants of the Eastern Cape

- Description: Understanding the mechanisms and adaptations that allow only certain species to thrive in the potentially stressful foredune environment requires a knowledge of the basic ecophysiology of foredune species. Ecophysiological measurements were conducted on the foredune pioneer species Arctotheca populifolia (Berg.) Norl., Ipomoea pes-caprae(L.) R. Br. and Scaevola plumieri (L.) Vahl. and showed significant differences among species with respect to the physiology associated with biomass production, water and nutrient relations. Differences related to CO₂ assimilation included differences in photosynthetic and respiratory rates, susceptibility to light stress and leaf and stem non-structural carbohydrate concentrations. These resulted in differences in primary production rates of shoots. Mechanisms leading to the differences in CO₂ assimilation among species included differences in stomatal behaviour, carboxylation efficiencies, efficiencies of utilisation of incident photosynthetic photon flux density (PPFD) and rates of ribulose-1,6-bisphosphate (RuBP) regeneration. Correlated with differences in photosynthetic capacity were differences in chlorophyll contents but not differences in leaf nitrogen content. Differences in interspecific stomatal behaviour resulted in significantly different transpiration rates which in combination with differences in assimilation rates resulted in differences in water-use efficiency. The absolute amounts of water transpired, although significantly different among species, were moderate to high in comparison with species from other ecosystems and were typical of mesophytes. Transpiration rates in combination with plant hydraulic conductances and soil water availability resulted in leaf water potentials that were not very negative and none of the investigated species showed evidence of osmotic adjustment. The volume of water transpired by each of the species per unit land surface area was estimated from the relationship between abiotic factors and plant water loss. These relationships varied among species and had varying degrees of predictability as a result of differences in stomatal behaviour between the three species. The water requirements of A. populifolia and S. plumieri were adequately met by the water supplied by rainfall and the water stored in the dune sands. It was therefore not necessary to invoke the utilisation of ground water or the process of internal dew formation to supply sufficient water to meet the requirements. However, I. pes-caprae despite its lower transpiration rates and due to its higher biomass, lost greater volumes of water per unit dune surface area than either A. populifolia or S. plumieri. This resulted in periods of potential water limitation for I. pes-caprae. Incident light was the most important determinant of leaf photosynthetic CO₂ assimilation and transpiration, particularly as a linear relationship between incident PPFD and atmospheric vapour pressure deficit (VPD) could be demonstrated. Whole plant photosynthetic production by S. plumieri was shown to be light limited as a result of mutual shading despite high incident and reflected PPFD occurring in the foredune environment. The leaf hair-layer of A. populifolia was shown to be important in reducing transmitted UV and hence reducing photoinhibition but it also caused reduced transpiration rates because of the thicker boundary layer and thus increased leaf temperatures. The nutrient content of above-ground plant parts of the investigated species were typical of higher plants despite the low nutrient content measured for the dune soils. With the possible exception of nitrogen the nutrient demand created by above-ground production was adequately met by the supply of nutrients either from sand-water or from aquifer-water transpired by the plants. Differences in the volumes of water transpired, and hence the quantity of nutrients potentially taken up via the transpiration stream, resulted in interspecific differences in above-ground plant macronutrient content. The reallocation patterns of nutrients differed both between the various nutrients measured and interspecifically. Standing biomass and the density of plants per unit land area was low in comparison to that of other ecosystems and was different among investigated species. This may be important in maintaining the adequate supply of resources (water, nutrients and light). As a result of the interspecific differences in biomass when production was expressed per unit land surface area the resultant productivity was not dissimilar among species. Productivity was high when comparisons were made with species from other ecosystems. No single resource (water, nutrients or light) could be identified as the controlling factor in the foredune environment and a combination of both resource stress and environmental disturbance are likely to be involved. Physiology, production, growth and growth characteristics conveyed certain adaptive advantages to these species in respect to both resource stress and environmental disturbance. Interspecific differences in these adaptations can be used to offer explanations for the observed microhabitat preferences of the three investigated species. Furthermore features common to all three species offer some explanations as to why these species and not others are able to inhabit the foredunes.

The effect of a soil-amending hydrogel on Eucalyptus grandis establishment practices in the Zululand forestry region

- Description: To ensure acceptable survival and initial growth of Eucalyptus grandis clonal and clonal hybrid material planted in the cool temperate and sub-tropical climates of Zululand (KwaZulu-Natal, South Africa) the planting seasons are currently restricted to the winter or summer months respectively. The Zululand coast (sub-tropical climate) experiences traditionally hot and humid summers and as a result Eucalyptus planting is largely restricted to the cool and dry winter months when survival is acceptable (90- 95%). In comparison, the Zululand interior (cool temperate climate) experiences moderately cooler summers followed by drier winters. As a result, the Eucalyptus planting season is reversed to that of the Zululand coast, with most of the planting taking place during the summer months with little or no planting occurring during winter. To ensure adequate transplant survival during and beyond these periods, transplants are planted with large volumes of water at a high cost. To determine whether it was possible to significantly reduce current water volumes at planting and therefore reduce costs, and to potentially extend the current planting seasons, two field trials were initiated during traditionally unsuitable planting periods (winter months for the Zululand interior and summer months for the Zululand coast). These trials were established near Kwambonambi on the Zululand coast and at Ntonjaneni in the Zululand interior. Five levels of water were combined with five levels of hydrogel and applied to the pit at planting in a 5 x 5 factorial treatment design for both trials. The tree variates of mortality, height, groundline diameter, crown diameter, corrected leaf surface index (LSIC) and corrected biomass index (BIC) were assessed at regular intervals until the final measurement dates (118 and 128 days after planting for the Ntonjaneni and Kwambonambi trials respectively). For the Kwambonambi trial, the response of transplant survival to the application water was highly significant (p<0,01) 128 days after planting, but not to the application of the hydrogel. Transplant survival nevertheless conformed to the silviculturally accepted norms of 90-95% using the hydrogel, thus water volumes could be significantly reduced without negatively affecting current survival standards. The lack of the expected response of significantly reduced transplant survival to increasing levels of hydrogel could possibly be attributed to the significant rainfall event (146 mm) that fell two days after trial initiation. Increasing levels of both water and hydrogel resulted in significantly enhanced growth (LSIC and BIC: p<0.01) for the final measurement date. For the Ntonjaneni trial, there was a significant (p<0,01) interaction between hydrogel and water, 118 days after planting. There were significant (p<0,01) differences between water only and all hydrogel treatments, with the hydrogel treatments performing significantly better. Optimum transplant survival for water only treatments was 50% using 4000 ml water while that for hydrogel treatments was 100% using 6 g hydrogel with 1000 ml of water and 12 g hydrogel with 2000 ml of water. - Description: To ensure acceptable survival and initial growth of Eucalyptus grandis clonal and clonal hybrid material planted in the cool temperate and sub-tropical climates of Zululand (KwaZulu-Natal, South Africa) the planting seasons are currently restricted to the winter or summer months respectively. The Zululand coast (sub-tropical climate) experiences traditionally hot and humid summers and as a result Eucalyptus planting is largely restricted to the cool and dry winter months when survival is acceptable (90- 95%). In comparison, the Zululand interior (cool temperate climate) experiences moderately cooler summers followed by drier winters. As a result, the Eucalyptus planting season is reversed to that of the Zululand coast, with most of the planting taking place during the summer months with little or no planting occurring during winter. To ensure adequate transplant survival during and beyond these periods, transplants are planted with large volumes of water at a high cost. To determine whether it was possible to significantly reduce current water volumes at planting and therefore reduce costs, and to potentially extend the current planting seasons, two field trials were initiated during traditionally unsuitable planting periods (winter months for the Zululand interior and summer months for the Zululand coast). These trials were established near Kwambonambi on the Zululand coast and at Ntonjaneni in the Zululand interior. Five levels of water were combined with five levels of hydrogel and applied to the pit at planting in a 5 x 5 factorial treatment design for both trials. The tree variates of mortality, height, groundline diameter, crown diameter, corrected leaf surface index (LSIC) and corrected biomass index (BIC) were assessed at regular intervals until the final measurement dates (118 and 128 days after planting for the Ntonjaneni and Kwambonambi trials respectively). For the Kwambonambi trial, the response of transplant survival to the application water was highly significant (p<0,01) 128 days after planting, but not to the application of the hydrogel. Transplant survival nevertheless conformed to the silviculturally accepted norms of 90-95% using the hydrogel, thus water volumes could be significantly reduced without negatively affecting current survival standards. The lack of the expected response of significantly reduced transplant survival to increasing levels of hydrogel could possibly be attributed to the significant rainfall event (146 mm) that fell two days after trial initiation. Increasing levels of both water and hydrogel resulted in significantly enhanced growth (LSIC and BIC: p<0.01) for the final measurement date. For the Ntonjaneni trial, there was a significant (p<0,01) interaction between hydrogel and water, 118 days after planting. There were significant (p<0,01) differences between water only and all hydrogel treatments, with the hydrogel treatments performing significantly better. Optimum transplant survival for water only treatments was 50% using 4000 ml water while that for hydrogel treatments was 100% using 6 g hydrogel with 1000 ml of water and 12 g hydrogel with 2000 ml of water. Different dietary energy and protein levels similarly had little or no effect on egg quality and composition. This study also assessed the carry-over effect of the nutritional regime in one year on the production in the following breeding season. Females fed diets as low as 7.5 MJ/kg ME in the previous breeding season produced significantly fewer eggs in the next breeding season, which resulted in lower chick production. Energy had no effect on the percentage infertile eggs or on the initial egg weight. Different levels of dietary protein had no effect on egg production, egg weight, fertility, hatchability and initial chick weight.

Therefore Microcystis aeruginosa strains were cultured under batch and continuous conditions to determine the effect of nitrate and phosphate concentrations and ratios on biomass and toxin production. Cultures were analysed with regards to internal nutrient stores, biomass production, nutrient depletion, photosynthetic efficiency and microcystin production. Results showed that microcystin production correlated to growth rate, photosynthetic efficiency and internal nitrogen stores and that an optimal N:P ratio was associated with microcystin levels, growth rate and photosynthetic efficiency. Results therefore led to the conclusion that the nitrogen, carbon, and phosphate balance within the cell is closely associated with microcystin production. Whether or not microcystin is produced to maintain this balance or produced as a function of this balance remains to be determined.

One of the toxins produced by cyanobacteria is microcystin, which is a potent hepatotoxin. Microcystin is taken up by bile acid transporters in the intestine and transported into the liver. After exposure to acute doses of microcystin, severe haemorrhage has been observed along with apoptotic and necrotic hepatocytes. The cytoskeletal structure of the hepatocytes is disrupted and oxidative stress is induced. Selenium, a known anti-oxidant, has been shown to induce increased activity of glutathione peroxidase. Glutathione peroxidase removes peroxides from cells protecting them from oxidative stress. This study set out to determine if selenium could play a role in preventing the damage to mice livers due to microcystin toxin. The protective role of selenium was explored in three main studies: in the first study, the ability of selenium to increase the survival time of mice exposed to a lethal dose of toxin was determined. In the second study the mice were exposed to sublethal chronic doses of toxin over 30 days. The ability of selenium to minimise liver damage under these conditions was determined. The final study investigated the mechanism of the protective effect of selenium. The results of the first study suggested that selenium could extend survival time. In the second study the selenium supplemented mice showed a reduction in the extent of the increase in liver weight and a decrease in the amount of lipid peroxidation induced compared to the mice that received only toxin. The histology of the selenium supplemented mice also showed a decrease in the severity and amount of morphological changes in the liver. The third study indicated that the protection shown by selenium might be mediated by an increase in the glutathione peroxidase (GPX) activity in selenium supplemented mice. This increase in GPX activity would increase the removal of the lipid hydroperoxides and prevent the damage they would cause in the cell. A further result indicated an increase in glutathione S-transferase in only the toxin control mice when compared to the selenium supplemented and control mice. ii In conclusion selenium offers protection against microcystin but further studies need to be done to provide statistically valid results to clarify the level of protection.

Testing involved the subjects having to perform 4 maximal turning efforts in 18 different conditions. These conditions were made up by using 6 different control designs in 3 varying positions. Subjects were required to attend two sessions, each approximately one hour long, in which nine randomised conditions were tested in each session. During these sessions, isokinetic responses: peak torque (Nm), total work (J) and average power (W); cardiovascular responses: heart rate (bt.min[superscript -1]) and blood pressure (mmHg); and psychophysical responses: RPE and discomfort, were observed. The results of the tests showed that in general significant differences were encountered for isokinetic, cardiovascular and psychophysical responses in relation to changes in the control design. However, significant differences were far less evident, and in most cases non existent, in relation to changes in the spatial orientation of the control types. The essence being that operator position with respect to the control is not as crucial as the control design.

Of particular interest was the selectivity and yield of the 1,4-dihydroxybenzenes. Two distinctly different methods were investigated in order to achieve this goal, acyloxylation and direct electrochemical hydroxylation. Acyloxylation is the process where radical cations generated at the anode undergoes nucleophilic attack by acetate anions. The resulting aromatic acetates so produced can then be hydrolysed to the phenolic compounds. Two nucleophile systems were considered in the investigation, acetates (acetoxylation) and trifluoro-acetates (trifluoro-acetoxylation). These investigations were conducted under a variety of conditions using phenol and phenyl acetate as starting materials. From the results it was, however, concluded that the acetoxylation of these aromatic compounds occurs in unacceptable product and current yields. Trifluoro-acetoxylation on the other hand showed promise, but due to the nature and cost of the reagents it was deemed to be an impractical process. Direct electrochemical hydroxylation: in which the radical cations produced at the anode undergoes nucleophilic attack by water producing the corresponding dihydroxybenzenes. - Description: Allan Waters, a communal area near Queenstown in the former Ciskei of the Eastern Cape Province of South Africa, have about 80 households who are involved in agriculture. These households were surveyed with two questionnaires. The first was at household level, focusing on the current socioeconomic situations and farming systems. Data were collected from sixty-three households in the village. The socio-economics questionnaire was divided into sections: demographic information, land and agricultural information, income and expenditure. In order to improve the livelihood of the households and to secure food production and food security, a situation analysis of the rural farmers was carried out using typology as a research tool. Typologies were built in order to analyze the diversity of production units (farms) or households (agricultural households). To define household typologies, factors such as land, source of income, modes of farming and composition of the family were used to identify six typology categories and to divide these into two main groups. The two main groups comprised of (a) livestock farming and (b) non-livestock farming or very little farming activities. The six types are as follows: Type 1: Poverty-stricken households Type 2: Newly arrived households Type 3: Households’ depending on old age pensions Type 4: Households’ main income from old age pensions and supported by substantial farming Type 5: Households’ revenue generated from business, wages and farming Type 6: Full time farming Five types had access to grazing and arable land for agricultural purposes, but type two had not yet been granted the rights to graze or the use of arable fields for agricultural purposes by the village. The source of income for the households in type one was made up of welfare grants and remittances. Types three and four, mainly depended on pensions when compared to the other four types. In the case of the wool farmers, the committee controlled activities such as mating of animals, grazing management, shearing, dipping of animals and wool sorting. The other tasks were left to the farmer himself. Wool data obtained for 2001 indicated that the average fleece weight was 3.88 kg greasy wool per sheep, with an A- to C- length. The fibre diametre of the wool in general fell in a medium class (21.1 micron). From the survey a number of projects were identified to assist the rural farmers in improving the livelihood of the households and in securing food production and food security.

- Description: Allan Waters, a communal area near Queenstown in the former Ciskei of the Eastern Cape Province of South Africa, have about 80 households who are involved in agriculture. These households were surveyed with two questionnaires. The first was at household level, focusing on the current socioeconomic situations and farming systems. Data were collected from sixty-three households in the village. The socio-economics questionnaire was divided into sections: demographic information, land and agricultural information, income and expenditure. In order to improve the livelihood of the households and to secure food production and food security, a situation analysis of the rural farmers was carried out using typology as a research tool. Typologies were built in order to analyze the diversity of production units (farms) or households (agricultural households). To define household typologies, factors such as land, source of income, modes of farming and composition of the family were used to identify six typology categories and to divide these into two main groups. The two main groups comprised of (a) livestock farming and (b) non-livestock farming or very little farming activities. The six types are as follows: Type 1: Poverty-stricken households Type 2: Newly arrived households Type 3: Households’ depending on old age pensions Type 4: Households’ main income from old age pensions and supported by substantial farming Type 5: Households’ revenue generated from business, wages and farming Type 6: Full time farming Five types had access to grazing and arable land for agricultural purposes, but type two had not yet been granted the rights to graze or the use of arable fields for agricultural purposes by the village. From the results obtained, it was clear that the excess of DIPEA and the temperature of the reaction both need to be high to obtain high yields. These theoretical results were confirmed by results obtained practically, where yields of up 82 % were obtained, but it became clear that even higher yields could be obtained since chromatographic results showed yeilds as high as 90 %. The mass balance of the MTSC synthesis showed a loss of approximately 30 grams per reaction. This loss may have an influence on the final yield. The effluent obtained during the synthesis of MTSC was investigated and a waste treatment protocol was established to reduce the high COD value of the MTSC effluent. The protocol consists of two steps used for the clean up of the effluent. The first being a cooling step; the effluent was cooled at 0oC to induce precipitation of a solid, consisting mostly of MTSC. The second step is a high pressure wet oxidation of the effluent with oxygen in a high pressure reactor. The remaining compounds in the effluent were oxidized, resulting in another precipitate, consisting mostly of sulphur. After the oxidation the COD value of the effluent was decreased by 98 % to a value of 0.4 %. The MTSC present in the precipitate obtained after cooling could be isolated and purified, to add to the yield of the synthesis. The sulphur obtained during the oxidation could also be isolated and reused, or sold to prevent it from contaminating the environment.

In relation to the (in)security of SADC member states, the thesis attempts to demonstrate that national integration of these states (i.e. the establishment of an amalgamated security community) has at least to accompany if not precede the establishment of a security community at the regional level (i.e. a pluralistic security community). Since threats to the security of SADC member states are mainly nonmilitary in nature, the 'realist' concept of security is broadened to include political, economic, societal and environmental aspects of security at different levels. Furthermore, Deutsch's concept of a security community is redefined in line with the 'new security thinking' and adapted to the situation in African states. Moreover, this thesis attempts to demonstrate that it is essential for SADC member states to become strong and socio-economically cohesive in order to improve their competitiveness in relation to developed states, especially in their ability to deal with internal and ransnational/regional threats to their security, which are (in)directly caused and perpetuated by the lack of national integration, inefficient state-making and underdevelopment - the sources of their weakness.

The Internet has had a major effect on news worldwide, and has contributed to sweeping changes in the news industry in all media. This study looks at the changes and the evolving strategy wrought by online news in the newspaper industry in two countries. In order to do this comparison, a model of the US experience has been constructed, using material published in the US academic and professional journalism press. Since there is no equivalent published material available dealing with the South African experience, interviews were conducted with staff at two newspaper companies (Johnnic and Naspers), and a broad historical overview was created for each company. These “narrative histories” were then compared with the US model, and areas of commonality and difference were highlighted and discussed. Several structural and national differences be tween the two countries were also raised and analysed. Finally, a conclusion as to how applicable the US model is to the South African experience is drawn, and suggestions are made for further study.

- Description: For the protection of the ecological Reserve in South Africa, the proposed introduction of compulsory toxicity testing in the licensing of effluent discharges necessitates the development of whole effluent toxicity testing. The elucidation of the effects of effluent on the local indigenous populations of organisms is essential before hazard and risk assessment can be undertaken. The limpet Burnupia stenochorias, prevalent in the Eastern Cape of South Africa, was chosen to represent the freshwater molluscs as a potential toxicity indicator. Using potassium dichromate (as a reference toxicant) and a textile whole effluent, the suitability of B. stenochorias was assessed under both acute and chronic toxicity conditions in the laboratory. In support of the toxicity studies, aspects of the biology of B. stenochorias were investigated under both natural and laboratory conditions. Using Principal Component and Discriminant Function Analyses, the relative shell morphometrics of three feral populations of B. stenochorias were found to vary. Length was shown to adequately represent growth of the shell, although the inclusion of width measurements is more statistically preferable. Two of the feral populations, one in impacted water, were studied weekly for 52 weeks to assess natural population dynamics. Based on the Von Bertalanffy Growth Equation, estimates of growth and longevity were made for this species, with growth highly seasonal. Age is not easily discerned from shell size. Egg laying occurred all year round, with early summer (peak egg lay), mid summer (a second, smaller peak in egg lay), and winter (limited presence of eggs) phases. In toxicity testing, consideration is given to the choice of the test organism based on age and sexual development. Consequently, the sexual development of B. stenochorias relative to shell length was determined with the aid of histological examinations of transverse sections of limpets, of all sizes, collected over one year. Limpets less than 3mm shell length were found to be immature in the development of the oocytes and spermatozoa, and were later chosen for acute toxicity tests. A laboratory diet was developed, for both culturing and maintaining of the limpets during toxicity tests; however, the diet requires optimisation. Under laboratory conditions, growth was linear, and individual fecundity highly variable. Successful methods for the collection of limpets from naturally occurring populations, and their acclimation to the laboratory were developed. Three B. stenochorias populations, representing different hydrological and water quality conditions, were compared to a laboratory population (maintained for three years) in their responses to the textile whole effluent and potassium dichromate. Under acute conditions, variability of mortality between limpet populations and between seasons was consistent with acceptable international standards. However, seasonal differences between feral limpets were apparent, with early summer limpets significantly more susceptible to both potassium dichromate and textile effluent than winter limpets. Although mortality occurred within the effluent at all concentrations, no 96 hour LC₅₀ values were obtained. The chronic toxicity effects of the textile whole effluent were assessed over the entire life cycle of B. stenochorias, based on survival, growth and reproductive effects. Lower concentrations of effluent (# 10%) gave greater variability of responses and toxicity than higher concentrations, with a 43 day LC₅₀ of 3.9% effluent. The No Observed Effect Concentrations for survival (over 43 days) were calculated in consecutive years as 0.1% and 1% effluent. Survival is considered a useful tool for determining toxicity endpoints using B. stenochorias. Limpet growth remained linear in effluent, with an apparent stimulation of growth at the 3-10% effluent concentration, confusing the toxicity and variability assessments. The possible addition of nutrients from the effluent points to either a potential inadequacy of the food quality provided in the chronic assessment, or the presence in the effluent of growth stimulants. Growth was also found to be too variable to allow adequate statistical conclusions about the toxicity of the effluent, although it is suggested that growth may be useful in the assessment of single compounds. Despite large individual variability in fecundity, statistical differences were discernible between effluent concentrations. The application of fecundity of B. stenochorias in hazard assessment therefore warrants further investigation. It was concluded that an assessment of textile whole effluent toxicity to B. stenochorias over an entire life cycle, and an F1 generation, is unnecessary. The development of the bucket/plastic bag method for both acute and chronic toxicity assessment of B. stenochorias was useful. In the final assessment of the usefulness of B. stenochorias as a toxicity indicator, toxicity endpoints were compared with those of the standard laboratory organism Daphnia pulex. Both in acute and chronic toxicity, B. stenochorias was found to be more sensitive. To achieve this objective theoretical models of competitive advantage and literature of industry clustering were identified. For the models on competitive advantage, Porter’s diamond was used for national competitive advantage and Porter’s model on the competitive environment, which affects the competitive advantage of a region. Other theories on competitive advantage of cities and regions were identified, leading up to the theory on the cluster approach. The literature mentioned was broken down and analysed using literature, from knowledgeable people in the automotive industry in the Eastern Cape, identified during the study. A questionnaire was developed to test the degree to which the catalytic converter industry cluster in the Eastern Cape is in agreement with the literature study. The empirical study obtained a strong concurrence with the literature study on national competitive advantage and the theory of clusters. This resulted in a strategy for the catalytic converter industry cluster to sustain competitive advantage and remain globally competitive.

For the models on competitive advantage, Porter’s diamond was used for national competitive advantage and Porter’s model on the competitive environment, which affects the competitive advantage of a region. Other theories on competitive advantage of cities and regions were identified, leading up to the theory on the cluster approach. The literature mentioned was broken down and analysed using literature, from knowledgeable people in the automotive industry in the Eastern Cape, identified during the study. A questionnaire was developed to test the degree to which the catalytic converter industry cluster in the Eastern Cape is in agreement with the literature study. The empirical study obtained a strong concurrence with the literature study on national competitive advantage and the theory of clusters. This resulted in a strategy for the catalytic converter industry cluster to sustain competitive advantage and remain globally competitive.

Of these free radicals, the O2–· and particularly the ·OH radical can induce cellular damage including lipid peroxidation. Thus, the use of antioxidants to prevent such damage induced by UV irradiation has received much attention recently. One such antioxidant, which has the potential to be incorporated into sunscreens, is the pineal secretory product melatonin. One of the concerns of using melatonin in sunscreens is its photostability. In the present study, we investigated the photostability of melatonin subjected to UV irradiation. In addition, we used liquid chromatography mass spectrometry (LC-MS) to identify the degradants and we also assessed the ability of the degradants to inhibit O2–· generation as well as lipid peroxidation in rat brain homogenate. The results show that UV irradiation of melatonin (0.1 mg/mL) using a 400-W lamp for 2 hr caused a significant decline of melatonin to 18% of its original concentration after 20 min, with the decline continuing until the melatonin concentration reaches zero at 120 min. The LC-MS results show that the degradants of melatonin are 6-hydroxymelatonin and N1-acetyl-N2-formyl-5-methoxykynurenamine (AFMK). These degradants were able to provide equipotent activity against potassium cyanide (KCN)-induced superoxide generation compared to non-irradiated melatonin. Thus, the study shows that although melatonin is rapidly degraded by UV irradiation, the degradants retain antioxidant activity, making melatonin a likely candidate for inclusion in sunscreens.

- Description: A module and an inoculation technique were developed that would allow for the efficient immobilization of Microcystis aeruginosa PCC7806 on a synthetic membrane. A variety of module types, membranes (ceramic, tubular polyethersulfone and externally skinless polyethersulfone capillary membrane), and methods of immobilization (adsorption, pressure filtration and a developed technique that involved drying a cell slurry on a membrane) were assessed. The morphological properties that affected the immobilization of Microcystis aeruginosa PCC7806, as well as the effects of immobilization upon cell morphology were assessed. Cells in the stationary growth phase, which had a well-developed extra-cellular polysaccharide layer and no gas vesicles, were optimal for immobilization. Microcystin production under immobilized conditions was assessed under different nitrate concentrations, light intensities, biofilm thickness and immobilization times. Additional work included assaying for Microcystin production of two airlift-grown cultures under a high light intensity and complete nutrient deprivation and the inoculation of a ceramic membrane. An immunological technique was used to elucidate where toxin production was greatest within a biofilm immobilized upon an externally skinless polyethersulfone capillary membrane. The externally skinless polyethersulfone capillary membrane was evaluated to assess homogeneity and the physical differences between membrane batches that led to the erratic, incomplete biofilm formation, as a biofilm of a constant thickness could not be immobilized. Microcystis aeruginosa PCC7806 was exposed to a variety of solvents in order to permeabilize the cyanobacteria, as that would have enabled a truly continuous extraction process for the metabolite. FDA hydrolysis had to be optimized in order to use it as an indicator of cell viability. In addition a single-step extraction of Microcystin was attempted using live bacteria. A capillary membrane module, containing the externally skinless polyethersulfone capillary membrane, inoculated using pressure filtration, was the most efficient combination to establish a biofilm. Cells that were no longer actively dividing and that lacked buoyancy displayed superior immobilization to cells that were actively dividing and buoyant. The immobilized cells did produce Microcystin but in much lower concentrations to cells grown in an airlift culture. Biofilms grown with a higher nitrate concentration, a lower biofilm thickness and a lower light intensity had a higher specific microcystin content, while biofilms with a higher nitrate concentration a lower light intensity and a longer growth period displayed the a greater toxin production per mm2 of membrane. Microcystin occurred at its highest concentration in cells just above the pore opening. The diffusion of nutrients occurred relatively quickly to the outside layers of the biofilm, with a true gradient being established laterally from these nutrient veins that were above the pores. Permeabilization of the cells proved unsuccessful, as cells that remained viable did not release the intracellular compound into the surrounding medium.

There were significant shifts in media industries: newspapers, broadcasting, cinema and telecommunications when governments begun re-regulating their air waves so as to permit private satellite transmission via both encryption and free-to-air, in addition to public service and private channels. In most societies where these changes have taken place, public service broadcasting has been threatened by the rapid rise of commercial institutions, resulting in stiff competition for audiences. This study set out to determine the extent to which commercialisation, in the era of liberalisation and commercialisation of media services in Uganda, has affected Radio Uganda’s programming. Using a combination of qualitative and quantitative methods of investigation, I have established that while Radio Uganda still maintains certain public service principles and values, programming policy has increasingly been changed by commercial considerations. This is shown by the recent rise of commercial programmes and a fall in education and developmental programmes. Limitations of finance and other resources have compromised the roles and character of public service radio programming. The majority of programmes currently on Radio Uganda are evidently geared to attract advertisers rather than serve the public interest. The study recommends, among other measures, that the licence fee be developed as a source of revenue for Radio Uganda. Secondly, government should inject more funding into public service broadcasting institutions to supplement other sources of income, before granting them autonomy. Thirdly, while advertising and sponsorship brings in a considerable amount of revenue, it should not take a central place that undermines the listener’s interest in radio programming. The Broadcasting Council should therefore map out solid policies that will systematically guide Radio Uganda in its programming in the new order.
